چکیده

At room temperature at stall, the flagellar motor of the bacterium Escherichia coli exerts a torque of ~1300 pN nm. At zero external load, it spins ~330 Hz. A robust method for studying the motor near zero load is reviewed here.

Thermal and solvent-isotope effects on the flagellar rotary motor near zero load.

In Escherichia coli, the behavior of the flagellar rotary motor near zero load can be studied by scattering light from nanogold spheres attached to proximal hooks of cells lacking flagellar filaments. We used this method to monitor changes in speed when cells were subjected to changes in temperature or shifted from a medium made with H(2)O to one made with D(2)O. In H(2)O, the speed increased w...

Resurrection of the flagellar rotary motor near zero load.

Flagellated bacteria, such as Escherichia coli, are propelled by helical flagellar filaments, each driven at its base by a reversible rotary motor, powered by a transmembrane proton flux. Torque is generated by the interaction of stator proteins, MotA and MotB, with a rotor protein FliG. The physiology of the motor has been studied extensively in the regime of relatively high load and low speed...

Limiting (zero-load) speed of the rotary motor of Escherichia coli is independent of the number of torque-generating units.

Rotation of the bacterial flagellar motor is driven by multiple torque-generating units (stator elements). The torque-generating dynamics can be understood in terms of the "duty ratio" of the stator elements, that is, the fraction of time a stator element engages with the rotor during its mechanochemical cycle. The dependence of the limiting speed (zero-load speed) of the motor on the number of...

The Effects of Stator Compliance, Backs Steps, Temperature, and Clockwise Rotation on the Torque-Speed Curve of Bacterial Flagellar Motor

Rotation of a single bacterial flagellar motor is powered by multiple stators tethered to the cell wall. In a " power-stroke " model the observed independence of the speed at low load on the number of stators is explained by a torque-dependent stepping mechanism independent of the strength of the stator tethering spring. On the other hand, in models that depend solely on the stator spring to ex...

Applying torque to the Escherichia coli flagellar motor using magnetic tweezers

The bacterial flagellar motor of Escherichia coli is a nanoscale rotary engine essential for bacterial propulsion. Studies on the power output of single motors rely on the measurement of motor torque and rotation under external load. Here, we investigate the use of magnetic tweezers, which in principle allow the application and active control of a calibrated load torque, to study single flagell...
